Epithalon Peptide: A Research Overview

Epithalon is a synthetic tetrapeptide (Ala-Glu-Asp-Gly) studied as a research compound in laboratory and animal models.
What Is Epithalon?
Epithalon (also written Epitalon or Epithalone, and known as the AEDG peptide) is a synthetic four-amino-acid peptide modeled on a component of the natural pineal-gland extract epithalamin. In the lab it is typically supplied as a lyophilized powder. The available data comes overwhelmingly from in-vitro (cell culture) and animal research; it has not completed large independent human clinical trials.
What the Research Describes
Mechanistic studies — in cells and animals — have investigated several overlapping pathways:
- Telomerase activity in telomerase-negative human fibroblast cell culture, where the peptide was associated with induced expression of the telomerase catalytic subunit, studied in vitro.
- Telomere elongation in human somatic cell culture, including reports of additional population doublings beyond the typical division limit.
- Neurogenic gene expression and protein synthesis in human stem-cell cultures (markers such as Nestin, GAP43 and Doublecortin).
- A proposed epigenetic / histone-binding mechanism examined via molecular modelling.
- Pineal and melatonin circadian regulation, studied in aged animal models and reported in the gerontology literature.
These describe mechanisms observed in laboratory and animal research — not demonstrated effects in humans.
A Note on the Current Evidence
- A large share of the Epithalon literature originates from a single research group (Khavinson and colleagues at the St. Petersburg Institute of Bioregulation and Gerontology), which limits independent replication.
- Much of the foundational work is published in Russian-language journals, making independent peer scrutiny and translation harder.
- The evidence base is overwhelmingly preclinical; there is a lack of large, independent, well-controlled human clinical trials.
Forms and Handling in Research
In a research setting Epithalon is handled as a lyophilized (freeze-dried) powder, commonly reconstituted with bacteriostatic water for laboratory use, stored cold and protected from light, and supplied with purity testing (e.g., HPLC) and lot documentation.
