What Kisspeptin-10 is
Kisspeptin-10 is a synthetic decapeptide corresponding to the shared C-terminal 10 amino acids of the peptides encoded by the human KISS1 gene — the same RF-amide-ending sequence found at the tail end of the larger 54-, 14-, and 13-amino-acid kisspeptin fragments (kisspeptin-54 is also referred to in older literature as metastin) [1]. All of these fragments, including the minimal decapeptide, have been reported to activate the G protein-coupled receptor GPR54 (now more commonly designated KISS1R) with low-nanomolar affinity [1].
In a research setting, Kisspeptin-10 is typically supplied as a lyophilized (freeze-dried) powder for reconstitution and in-vitro or in-vivo investigation. It is supplied for laboratory research use only and is not for human consumption.
Pathways published research has investigated
Published studies — spanning receptor pharmacology, human genetics, and controlled dosing trials in both men and women — have investigated Kisspeptin-10 and the broader kisspeptin/GPR54 system across several biological pathways. These are descriptions of what researchers have studied, not statements of effect in humans:
GPR54/KISS1R signaling and the reproductive axis. The kisspeptin family was first characterized in 2001 as the natural ligand for the previously orphan receptor GPR54, isolated from human placental tissue and shown to trigger calcium mobilization and other downstream signaling in cells expressing the receptor [1]. Two years later, human genetics research linked inactivating mutations in GPR54 to idiopathic hypogonadotropic hypogonadism — a failure to enter puberty — establishing the receptor, and by extension its kisspeptin ligands, as a central regulator of the hypothalamic-pituitary-gonadal (HPG) axis [2].
LH pulsatility in men. The first-in-human study of Kisspeptin-10 specifically administered intravenous bolus doses (0.01–3.0 µg/kg) and continuous infusions to healthy men. A lower-dose infusion (1.5 µg/kg·h over 9 hours) was reported to increase LH pulse frequency from approximately 0.7 to 1.0 pulses per hour and to raise the proportion of LH released as pulsatile secretion, while a higher-dose infusion sustained elevated LH for over 22 hours without observed tachyphylaxis in that timeframe [3].
Hypogonadism context. A separate study administered Kisspeptin-10 to men with type 2 diabetes and mild biochemical hypogonadism, reporting increases in serum testosterone and LH secretion in that population — a step toward understanding the peptide's studied activity outside of healthy young volunteers [4].
Cycle-dependent gonadotropin response (kisspeptin-54). Related human research using the longer kisspeptin-54 fragment reported that subcutaneous injection increased plasma LH in women at every phase of the menstrual cycle tested, with the largest response during the preovulatory phase and a markedly smaller response during the follicular phase [5].
Oocyte maturation trigger (kisspeptin-54). In a clinical fertility research setting, a single subcutaneous dose of kisspeptin-54 was used to trigger oocyte maturation in women undergoing IVF, with egg maturation observed at each of four doses tested and fertilization proceeding in the large majority of the 53 participants studied [6].
Sexual and emotional brain processing (kisspeptin-54). Using functional MRI, a randomized placebo-controlled crossover study in 29 men reported that kisspeptin infusion increased limbic-region brain activity (including the amygdala and hippocampus) in response to sexual and couple-bonding imagery, alongside changes in self-reported mood measures during the infusion window [7].
The state of the literature
Kisspeptin-10 and its longer relative kisspeptin-54 have an unusually deep human-dosing literature for research peptides of this kind — first-in-human pharmacology in men, dosing across the female menstrual cycle, an IVF fertility trial, and neuroimaging studies have all been published in peer-reviewed journals [3, 4, 5, 6, 7]. That is a meaningfully different evidentiary picture than compounds studied only in animal models.
It is important to characterize the limits accurately, though. Chronic or repeated kisspeptin-54 dosing has been reported to produce tachyphylaxis (a diminishing hormonal response with continued exposure) in some populations, including women with functional hypothalamic amenorrhea, even though a separate study found twice-daily dosing over one week did not abolish menstrual cyclicity in women with normal cycles [8]. No kisspeptin-based compound — Kisspeptin-10 included — has been approved by the FDA or any regulator as a drug for any indication, and none is a nutritional supplement. The IVF-trigger and reproductive-hormone research described above remains investigational, and the neuroimaging findings on mood and sexual/emotional brain processing are early-stage mechanistic observations, not an approved treatment for any sexual or mood-related condition.
This is exactly why credible discussion of Kisspeptin-10 stays in the research register — "studies have investigated," "the trial reported," "researchers observed" — rather than making claims about what the compound does for a person.
How researchers handle it
In laboratory use, Kisspeptin-10 is reconstituted from lyophilized powder, commonly with bacteriostatic water, and handled under standard research conditions. Material supplied for research should carry a Certificate of Analysis confirming HPLC-verified purity so that what is being studied is well-characterized.
